The sight of Peter wearing the symbiote suit implies that the symbiote will play a significant role in gameplay. However, as Venom has been confirmed to be a villain, it is evident to fans that Peter will not be bonded with the symbiote for the entirety of the game.
These revelations, along with other details shared by Insomniac, hint at the direction of the sequel. Each trailer seems to reveal the main focus: Harry's return from stasis and his collaboration with Peter to potentially find a cure for his illness, while Kraven pursues his chosen targets.

While Mr. Negative did serve as the primary villain for a considerable period, his prominence was swiftly surpassed in the third act when Otto Octavius, known as Doctor Octopus, publicly embraced his villainous persona. Unfortunately, this crucial development had unsurprisingly been leaked prior to the game's release.
Insomniac may have already revealed an important part of the villain or storyline in Marvel's Spider-Man 2, possibly to prevent spoilers and give fans a glimpse of what to expect. While keeping certain details sacred until players explore them is crucial, hiding too much like in The Last of Us Part 2 could negatively impact fans' expectations. However, if players have already been introduced to the main antagonists and the gameplay style in the sequel for each Spider-Man, it could be a positive aspect. Revealing too many secrets or hidden storylines could potentially clutter the packed sequel with content.
Introducing another antagonist could harm the overall plot, and concealing them solely to preserve the mystery of at least one villain may not be as fulfilling as fully unveiling and developing them within the story itself.
