The leading role in Gerwig's Barbie is portrayed by Margot Robbie, as she portrays Mattel's iconic doll who embarks on an adventure outside of Barbieland after experiencing unexplainable events that make her question her life. Despite the impressive cast, Gerwig and Robbie had initially mentioned the possibility of Chalamet and Saoirse Ronan, the lead in Lady Bird, making cameo appearances. Unfortunately, scheduling conflicts prevented this from happening. During an interview with Hollywood First Look, Gerwig discussed an occasion during production when Chalamet visited the set. Upon the completion of filming for Barbie, a remarkable ensemble of well-known personalities from the entertainment industry had already assembled. This star-studded cast comprised comedic legends such as Will Ferrell and Kate McKinnon, as well as esteemed acting icons like Helen Mirren. These individuals' familiar faces are sure to be recognized by viewers, thanks to their notable performances in other celebrated productions. However, it is noteworthy that Chalamet and Ronan's brief appearances in Barbie were not the only ones that almost materialized. Several other actors were on the verge of joining this impressive lineup, but various circumstances ultimately prevented them from doing so.

Gal Gadot was initially considered for a role in Barbie, but her busy schedule prevented her from participating. The casting directors, Lucy Bevan and Allison Jones, revealed several other potential Kens including Bowen Yang, Dan Levy, and Benn Platt. Jonathan Groff was also considered to portray Barbieland's only remaining Allan, but the role ultimately went to Michael Cera. While Helen Mirren provided her voice for the film, there were plans for her to have a physical appearance in a post-credits scene featuring the pregnant doll Midge, played by Emerald Fennell.

With the success of Barbie and talks of a possible sequel, many actors have expressed interest in joining the cast. Cillian Murphy, for example, has shown his desire to portray Ken. If Gerwig decides to direct the Barbie sequel, there might be another opportunity for Timothée Chalamet to return to Barbieland once again.
